EICOSANOIDS AND RELATED COMPOUNDS – AN INTRODUCTION Structures and Key Enzymes

چکیده
The term eicosanoid is used to embrace biologically active lipid mediators (C20 fatty acids and their metabolites), including prostaglandins, thromboxanes, leukotrienes and other oxygenated derivatives, which are produced primarily by three classes of enzymes, cyclooxygenases (COX-1 and COX-2), lipoxygenases (LOX) and cytochrome P450 epoxygenase. The key precursor fatty acids are 8c,11c,14c-eicosatrienoic (dihomo-γ-linolenic or 20:3(n-6)), 5c,8c,11c,14ceicosatetraenoic (arachidonic or 20:4(n-6)) and 5c,8c,11c,14c,17c-eicosapentaenoic (20:5(n-3) or EPA) acids (see our web page on ‘polyunsaturated fatty acids’). However, it is now impossible to discuss these compounds and their biological activities properly without also considering related products produced by non-enzymic means (isoprostanes), and the docosanoids (resolvins) derived from 4c,7c,10c,13c,16c,19c-docosahexaenoic acid (22:6(n-3) or DHA). Nor can analogous plant products, such as the jasmonates and other oxylipins derived from 9c,12c,15coctadecatrienoic (α-linolenic or 18:3(n-3)) acid, be ignored. It is noteworthy that the precursor fatty acids belong to both the omega-6 and omega-3 families.

متن کاملThe 2-Series Eicosanoids in Cancer: Future Targets for Glioma Therapy?
The 2-series eicosanoids are structurally related lipid-soluble hormones synthesized by cyclooxygenase enzymes from arachidonic acid. These compounds have well-established roles in the inflammatory response and the coagulation cascade. More recently, the eicosanoids have garnered attention for their potential roles in cancers of the lung, colon, breast, and brain. In this paper, we review the c...
متن کاملEicosanoids Derived From Arachidonic Acid and Their Family Prostaglandins and Cyclooxygenase in Psychiatric Disorders
Arachidonic acid (AA)-derived lipid mediators are called eicosanoids. Eicosanoids have emerged as key regulators of a wide variety of physiological responses and pathological processes, and control important cellular processes. AA can be converted into biologically active compounds by metabolism by cyclooxygenases (COX). Beneficial effect of COX-2 inhibitor celecoxib add-on therapy has been rep...
متن کاملProstaglandin endoperoxide synthase: why two isoforms?
Prostaglandin endoperoxide synthase-1 [prostaglandin G/H synthase-1 (PGHS-1)] and PGHS-2 are key enzymes in the conversion of arachidonic acid to prostaglandins and other eicosanoids. We refer to these isoforms as cyclooxygenase-1 (COX-1) and COX-2 in this review. This brief review focuses on recent developments in the study of these enzymes. Alterations in the expression levels of COX-2 result...
متن کاملThe biology of prostaglandins and related eicosanoids in invertebrates: cellular, organismal and ecological actions
Prostaglandins and related eicosanoids are oxygenated metabolites of C20 polyunsaturated fatty acids. These compounds have been detected in species representing all major animal phyla. The significance of eicosanoids lies in two broad areas of animal biology. In one, eicosanoids are involved in regulation of many cellular events. In the other, eicosanoids facilitate certain ecological interacti...
